Conventional conjugation technologies used for the development of ADCs exploited the superficial lysine residues that were inherent to the native antibody structure, or the inter-chain cysteine residues, for drug-linker attachment. Over time, the evolution of recombinant DNA technology and advances in conjugation techniques, have enabled researchers to significantly improve the bioconjugation process. In fact, next generation conjugation technologies use artificially integrated lysine / cysteine residues in order to improve the DAR of the resulting conjugate. Additionally, there are multiple technologies that claim to offer site-specific conjugation.
