New book by William J. Walsh, Ph.D., reveals how ion channel gene weaknesses and DNA damage drive the mania–depression cycle.

Based upon recent advances in neuroscience and genetics, this information has led to an explanation for mania/depression switching, new treatment opportunities, and an advanced theory of bipolar disorder.
More than nine million Americans are diagnosed with bipolar disorder, a leading cause of suicide, unemployment, and loss of human potential. For decades, its biological origins have remained elusive—until now. Dr. Walsh's multi-year study, presented at a recent Society for Neuroscience conference, reveals that bipolar disorder's strong genetic predisposition is caused by two coexisting factors:
- A genetic weakness in the team of 400+ collaborating ion channel genes, and
- A genetic tendency for accelerated DNA damage.
According to Dr. Walsh, these impairments usually are not sufficient to produce the illness during childhood or adolescence. However, several years of relentless free-radical assault and DNA damage eventually cause hyperactivity of major neurotransmitters and onset of mania. Serotonin neurons experience the greatest damage due to non-stop flooding of external potassium ions, leading to severe hypoactivity that causes the switch to depression. Recovery of serotonin function occurs automatically due to reduced potassium ion outflow and surviving clearance mechanisms that gradually eliminate potassium ion flooding.
